Ernst Stavro Blofeld:

Bond comes face to face with his arch enemy, Blofeld, in the film. Ernst Stavro Blofeld is the head, "Number One", of the SPECTRE crime organization. Played by actor Anthony Dawson (well below his head anyway) who played Professor Dent in Dr. No and dubbed by Eric Pohlman, Blofeld proves to be the most mysterious and formidable foe of the entire series.
We get our first glimpse of Blofeld when he calls in his agents, Klebb and Kronsteen, to question the status of the plan to obtain the Lektor. We see only his neck down and he's always seen stroking a fluffy white cat. Probably the best villain the series will ever see.
Rosa Klebb:
Lotte Lenya is superbly cast as the sick and twisted former Russian Colonel Rosa Klebb. Lenya is perfect for the role, as she can look menacing enough and her acting talent is more than satisfactory. Interestingly enough, most cast and crew associated with the film will attest that Lenya was an extremely kind lady in real life, the total opposite of her character. This only goes to further demonstrate her acting ability.
Klebb's character was especially controversial at the time due to the underlying lesbian tendecies in the film (shown in her attitude toward Romanova). Klebb is in charge of executing Kronsteen's plans and choosing the right people for the job. Ultimately she fails and ironically dies at the hands of Tatiana Romanova.
Kronsteen:
Polish actor Vladek Sheybal plays Kronsteen, Czech chess prodigy and SPECTRE agent. Sheybal had to be persuaded by Connery to take the role but later admitted that he was glad he had done it. Sheybal does a nice job at playing Kronsteen, but the character isn't developed as he doesn't have much screen time (which isn't neccesarily bad).
Kronsteen is the head of planning for SPECTRE and develops a "foolproof" plan to eliminate Bond and capture the Lektor. He pays dearly for his failure when he's killed with the poison from Morzeny's blade concealing boot.
Donald "Red" Grant:
Convicted murderer Donald "Red" Grant escaped Dartmoor prison to become one of SPECTRE's most skilled assasains. Robert Shaw skillfully plays Grant, which results in the villain becoming one of the most memorable in the series.
Grant boards the Orient Express to kill Bond and Tania and later poses as the reinforcement Bond requested. Bond spots the intrusion when Grant orders red wine with fish (apparently something an Englishman would never do) and drugs Tatiana to sleep to keep her out of the way while he kills Bond. The fight between Bond and Grant in the small train cabin is one of the highlights of the the film.
Morzeny:
Actor Walter Gotell plays SPECTRE head of traning/assasain Morzeny. Although we see little of Morzeny, Gotell's performance is commendable. Gotell would later return as General Gogol throughout the Moore and Dalton eras.
Morzeny leads Klebb around the SPECTRE island training facility and kills Kronsteen when he fails. Morzeny dies along with other SPECTRE killers in an explosion Bond sets off during the boat chase near the end of the film.
